I can't compile a webassembly empty project in my home PC but I can in the PC of my office.
I have Unity 5.6.0f3 in both computers, both run Windows 10.

    Bug report:
    https://fogbugz.unity3d.com/default.asp?899225_3v2a486js2ej5oib

Seems to be related with a path problem or windows permissions, I tried opening Unity as administrator but it doesn't work. Also tried creating the project in the root C:/projectName (a path without spaces) but nothing.
When trying to compile I get this error:

The only strange thing in this computer is that I did a partition to be used as temp, see the enviroment variables here where TEMP = F:\:

    EDIT:

Confirmed! It compiles fine when I change the TEMP and TMP variables to a folder inside C:\, I tried different folder names and it works (as long as it is inside C) like this:

I tried this and moved my user temp folders to another partition but can successfully build an asm and wasm build.

Can you file a bug report as described https://unity3d.com/unity/qa/bug-reporting here. And include info like the path to your project and to your Unity install.

I changed my temp folder to F:\Temp and I could build, so the problem is when your temp folder is a root path like: F:\.
I know that this sounds like a not important bug, but behind this bug can be the one of the other post that roka says.

Try this configuration please: TEMP and TMP = Partition root

Got it now and will be passing the bug on to the development team.
