I'm also interested in this topic. I tried all the mentioned key combinations but no flipping or rotation.

To be on the same page:
    1. I mark my tiles in the tiles palette
    2. Select the brush tool (from the palette)
    3. While using the brush with the selected tiles (in the grid of the scene view) I tried the key combinations for rotation and flipping but nothing happend

    Has anybody an idea?
(Besides: I'm using a german QWERTZ keyboard if there might be some shortcut translation problems)

Check the key in Unity preferences. [Edit -> Preferences -> "Keys" tab -> Grid Painting / Flip X]

Marvelous. Thank you very much @LCStark this hint solved my problem :):):).
To flip x it was set to "shift + leftbracket" by default but this didn't worked so I customized it to "shift + x" and now everything works just fine :D.
